Custom Peptide Synthesis

General overview:

We believe that quality products save you time. Our peptide synthesis know-how have been in place since 2001. To date we have synthesized over 1,500 peptides (5-50 mg scale) for academic research and pharmaceutical companies. Peptides are normally shipped within 2 weeks from the reception of your order.

Sequence analysis:

Feasibility evaluations are conducted on all peptide sequences prior to synthesis. Using advanced software and based on our own internal database of difficult to synthesize sequences, a determination is made regarding the complexity of the synthesis. For antibody production we also include antigenic index (Jameson-Wolf) and surface probability evaluations. Should any of these parameters be too low you will be informed prior to the acceptance of your order.

Chemistry:

Inbiolabs uses fully automated instruments for the production of custom peptides. Peptides are synthesised using Boc or Fmoc chemistry and purified by HPLC using reverse phase C4 and C18 columns.

Purification and quality control:

All peptides are analyzed by MALDI Mass Spectrometry to determine the molecular weight of the synthesized peptide, free of charge. MALDI is an excellent method for determining the presence of the desired product, but an inadequate method for the determination of purity. Due to the variant efficiencies of peptide ionization in the MALDI instrument, peak intensities in MALDI spectra are not necessarily quantitative. For this reason all non-crude peptides at Inbiolabs are analysed by Reverse-Phase HPLC (RP-HPLC) to determine purity.
Precipitated peptides are analysed on a RP-HPLC instrument equipped. Peptides that meet or exceed the purity requirements are then lyophilized, packaged and shipped to the customer at room temperature by overnight dispatch.

Customer will receive*:

  • 20 mg of crude peptide (55-75% pure) // 5, 10, 15 or 20 mg of purified peptide (>95%)
  • MS and HPLC analyses data

Available also**:
  • other amounts (milligrams to gram scale)
  • different purity
  • peptides shorter than 10 amino acids and longer than 34 amino acids
  • multiple modifications eg
               phosphopeptides (phosphorylated Tyr, Thr, Ser)

    cyclic peptides
               peptides with D-amino acids
               production of polylysine matrices
               peptide termini modification (acetylation, amidation)
conjugations to fluorescent tags and dyes (eg FITC, Alexa, ATTO532, Cy3B)
bioconjugations (eg biotin, HRP, KLH, BSA, OVA, cell-permeable peptide sequences)
